چکیده
conclusions community acquired (ca)-mrsa is becoming a public health concern in many parts of the world, including asian countries. the variable prevalence of luk-pv-positive mrsa isolates in different regions and their rather high frequency in pneumonia necessitate the application of rapid diagnostic methods such as real-time pcr to improve treatment effectiveness. materials and methods in total, 284 staphylococcus isolates, identified by phenotypic methods from clinical samples of shahrekord university hospitals, shahrekord, iran, were tested for nuc, meca, and pvl genes by taqman real-time pcr. the antibiotic susceptibility patterns of pvl-containing mrsa strains were determined via the disk diffusion method. background a diversity of virulence factors work together to create the pathogenicity of staphylococcus aureus. these factors include cell surface components that promote adherence to surfaces as well as exoproteins such as panton-valentine leukocidin (pvl), encoded by the luk-pv genes, that invade or bypass the immune system and are toxic to the host, thereby enhancing the severity of infections caused by methicillin-resistant staphylococcus aureus (mrsa). results in total, 196 isolates (69%) were nuc positive (i.e., s. aureus); of those isolates, 96 (49%) were meca positive (mrsa). eighteen (18.8%) of the 96 mrsa positive and 3 (3%) of the 100 methicillin-susceptible staphylococcus aureus (mssa) strains were pvl positive. pvl-positive mrsa strains were mostly recovered from tracheal specimens. eight pvl-positive mrsa strains were resistant to all the tested antibiotics except vancomycin. a significant correlation (p = 0.001) was found between the meca positivity and the presence of luk-pv genes. objectives the aim of this study was to determine the frequency of pvl-positive mrsa strains by real-time pcr and their antibiotic susceptibility patterns by phenotypic test.

متن کاملStaphylococcus aureus isolates carrying Panton-Valentine leucocidin genes in England and Wales: frequency, characterization, and association with clinical disease.
Staphylococcus aureus isolates carrying the genes that encode for Panton-Valentine leucocidin (PVL), a highly potent toxin, have been responsible for recent outbreaks of severe invasive disease in previously healthy children and adults in the United States of America and Europe. To determine the frequency of PVL-positive isolates sent to the Staphylococcus Reference Unit (United Kingdom) for ep...
متن کاملA brief history of Staphylococcus aureus Panton Valentine leucocidin
The history of Panton Valentine leucocidin (PVL), one of the toxins of Staphylococcus aureus, began in 1894, 24 years after Sir Alexander Ogston and Louis Pasteur had reproduced human pyemic infections in mice and rabbits by inoculating pus or pus-derived cultures (11,14). The torch of PVL research was then handed down over the decades between small groups of scientists fascinated by S. aureus ...
متن کاملGenetic characterization of Staphylococcus aureus isolates carrying Panton-Valentine leukocidin genes in Bangladesh.
To assess the spread and genetic characteristics of Panton-Valentine leukocidin (PVL) gene-carrying Staphylococcus aureus in Bangladesh, we investigated 59 strains (49 isolates from clinical specimens and 10 isolates colonized in the nasal cavities of medical staff), including 26 methicillin-resistant S. aureus (MRSA) strains. The PVL gene was detected only in methicillin-susceptible S. aureus ...
متن کاملPanton-Valentine Leukocidin Genes in Staphylococcus aureus
To the Editor: The pathogenicity of Staphylococcus aureus depends on various bacterial surface components and extracellular proteins. However, the precise role of single virulence determinants in relation to infection is hard to establish. The frequent recovery of staphylococcal isolates that produce leukocidal toxins from patients with deep skin and soft tissue infections, particularly furuncu...
متن کاملMolecular characterization and panton-valentine leucocidin typing of community-acquired methicillin-sensitive Staphylococcus aureus clinical isolates.
Limited comprehensive molecular typing data exist currently for Panton-Valentine leucocidin (PVL)-positive, methicillin-sensitive Staphylococcus aureus (PVL-MSSA) clinical isolates. Characterization of PVL-MSSA isolates by multilocus sequence typing (MLST) and spa typing in this study showed a genetic similarity to PVL-positive, methicillin-resistant S. aureus (PVL-MRSA) strains, although three...
